Separation and analysis of hemoglobin variants with PolyCAT A
PolyCAT ATM ia the gold standard worldwide for analysis of hemoglobin variants by cation-exchange HPLC. The most common variants can be separated in 3.5 minutes with good quantitation and with a turnaround time between samples of 4 - 4.5 minutes. This includes quantitation of Hb A1c for diagnosis of diabetes. The examples below were obtained with the widely-used 35 x 4.6 mm column packed with the 3-μm, 1500-Å pore version of PolyCAT A (item 3.54CT0315).
These variants were analyzed with a slightly longer column to improve separation.

Column: PolyCAT A, 35 x 4.6 mm, 3-μm, 1500-Å (item 3.54CT0315)
Solvent A: 20 mM bis-Tris + 2 mM KCN, pH 6.8
Solvent B: 20 mM bis-Tris + 2 mM KCN + 200 mM NaCl, pH 6.8
Gradient: 0-8 min, 2-25% B; 8.1-18 min, 25-100% B.
Flow rate: 1.5 ml/min.
